It is important that the RT buffer provided in the NEBNext Single Cell/ Low input RNA library prep kit and module remains a homogeneous solution. This means that this buffer needs to be vortexed well prior to each use for optimal reverse transcription efficiency. Vortexing is more efficient than pipette mixing because it makes reverse transcription efficiency higher.
